Robert Neville (Will Smith) is the last man standing in New York City—and possibly the world. A modified measles virus, originally intended to cure cancer, has wiped out most of humanity. The survivors have been turned into "Darkseekers"—nocturnal, zombie-like creatures with a thirst for blood. Neville, a virologist, spends his days scavenging, sending out radio broadcasts, and working on a cure in his basement lab, accompanied only by his dog, Samantha.
